Abstract
The pegagan (Centella asiatica (L.) Urban) contents triterpenes such as asiaticoside, madecassoside, asiatic acid, and madecassic acid that can speed up the formation of collagen in the wound healing process. Purpose. To know the optimal dose of ethanolic extract of pegagan with concentration 40%, 50%, 60% for density of collagen in the wound healing process in Sprague Dawley rats’ skin. Methods. This research is pure experimental methods with in vivo study. The subject of this research are 36 male Sprague Dawley aged 2 month old were divided into 4 groups randomly, each group consisting of 9 rats. A wound was made on rat’s back 2 cm long and 0,1 cm depth. One hundred μl of aquades, 40%, 50% and 60% of pegagan extract were given on the wound of each treated groups twice a day at intervals of 8 hours. The evaluation of all groups of rats were made on 3, 7, 12 days after being wounded. Histologic preparations were made with Mallory staining. Density of collagen fibers were observed microscopically in blue light colors. Result. The result showed that the pegagan extract with concentration 60% can accelerate the process of forming collagen fibers. Kruskal Wallis test showed statistically significant difference (p <0.05) compared to the control group, the group with 40%, 50%, and 60% application of extract in the process of wound healing at day – 3, 7, and 12. Results of Mann - Whitney test day - 3 and 7 showed significant difference between the control group and 60% group of pegagan extract, group 40% and group 60% of pegagan extract, group 50% and group 60% of pegagan extract. Mann-Whitney test on day - 12 showed the results that there are significant differences between the control group and the group of 60% pegagan extract, the group of 40% and the group of 60% pegagan extract. Conclusion. Ethanolic extract of pegagan with 60% concentration is the optimal dose for collagen formation in the wound healing process in Sprague Dawley’s skin.
